1927年9月，秋收起义之后，雷刚（马永安 饰）在湘赣边界领导的农民自卫军由于缺乏有组织的领导，加之人员组成复杂，经三起三落后，濒临灭亡。次年春天，党组织派来的党代表柯湘（杨春霞 饰）在来杜鹃山的途中被毒蛇胆的靖卫团逮捕，雷刚带人勇劫法场。回山后，针对雷刚让人误打田大江（高牧坤 饰）一事，柯湘对队员们进行阶级教育和政策教育。部队打土豪、分田地，开仓放粮，赢得群众爱戴。叛徒温其久（王忠信 饰）勾结毒蛇胆抓走杜妈妈（刘桂欣 饰），引诱雷刚下山营救。头脑简单的雷刚在叛徒的挑唆下冒然下山，致使自卫军遭受重大损失，自己则被捕入狱。母子狱中相见，面对杜妈妈对其轻率行为的批评，雷刚悔恨不已。危急关头，柯湘沉着冷静，带尖刀班雨夜奇兵营救...... 本片根据20世纪60年代同名话剧改编。
